FOXO4-DRI is a synthetic D-retro-inverso peptide engineered to disrupt the interaction between the FOXO4 protein and the tumor suppressor p53. In senescent cells, FOXO4 is thought to keep p53 in the nucleus and prevent cell death; blocking this interaction is proposed to release p53 to trigger apoptosis, selectively eliminating senescent cells. The evidence is entirely preclinical, based on cell and mouse studies. Because it acts on p53, a critical tumor-suppressor protein, its safety in humans is unknown.
