P21 (P021) is a synthetic adamantane-modified tetrapeptide derived from the active region of human ciliary neurotrophic factor (CNTF). In preclinical work it is described as a neurogenic and neurotrophic compound that enhances dentate gyrus neurogenesis by inhibiting LIF signaling and increasing BDNF expression, and that reduces abnormal tau hyperphosphorylation via BDNF-mediated decreases in GSK-3 beta activity. It is reported to be blood-brain-barrier permeable and orally available in animal models. It remains an investigational compound without human safety or efficacy data.
