Retatrutide is an investigational single molecule that acts as an agonist at three receptors: GIP, GLP-1, and glucagon. The added glucagon-receptor activity is thought to increase energy expenditure alongside the appetite and glycemic effects of the incretin pathways. It is in late-stage clinical development and is not yet approved.
